Ziel des Praktikums ist die Vorstellung eines Verfahrens, das es erlaubt, die für die Sichelzellanämie verantwortliche Punktmutation in der ß-Kette des Hämoglobins zu erkennen.
Dazu wird das ß-Globin-Gen einer Kontrollperson und das eines Patienten mit Sichelzellanämie mit einer Restriktionsendonuklease gespalten, die ihre Nukleotiderkennungsregion in dem mutierten Abschnitt der DNA hat.
Das entstehende Spaltmuster der normalen DNA ist deshalb verschieden von dem der mutierten DNA ( Restriktions-Fragment-Länge... ).

For this purpose, the ß-globin gene from a control person and that of a patient with sicklecell anaemia are split with a restriction endonuclease which has its nucleotide recognition in the mutated section of the DNA.
The split sample of the normal DNA which arises therefore differs from that of the mutated DNA ( restriction fragment length polymorphism ).
