Mit der Verfügbarkeit der reversen Genetik für Influenza A Viren, und der damit verbundenen Möglichkeit der direkten Manipulation des Genoms der Erreger haben sich die Möglichkeiten zur Untersuchung der molekularen Mechanismen der Infektion als Vorbedingung der Entwicklung effektiver Bekämpfungsstrategien verbessert.
Wir haben die für typische Vertreter der 16 Subtypen des Hämagglutinins kodierenden Sequenzen in den Expressionsvektor pHW2000 ( Hoffmann et al., PNAS 2000 ) kloniert. Diese 16 unterschiedlichen Hämagglutinine wurden mittels in vitro Mutagenese jeweils mit einer polybasischen Spaltstelle ausgestattet.
